For everyone saying Ledger in TDK, please check out this Tom Waits interview. I'm not saying it takes away from his performance, but this was an impression compared to an organic character.
Some other greats not mentioned...
Benecio Del Toro as Oscar Acosta in Fear and Loathing
Naomi Watts - Mulholland Drive
Philip Seymore Hoffman and Meryl Streep- Doubt
Val Kilmer is an interesting actor. He knocked Tombstone and The Doors out of the park, but then has horrible roles.

My dad always says the greatest performance he's seen was Dustin Hoffman in Midnight Cowboy. Hoffman was also so great in Tootsie -- he nailed playing a woman.
I've always held Philip Seymour Hoffman's role in Boogie Nights in high regard. Everyone got to be cool and sexy in that movie, and he was just a clumsy, awkward, closeted gay guy.
