There are a dozen other legit steak houses that can be found in other cities as well, but you definitely want to find a steakhouse that only exists in that city. And for that, Keen's is your answer.
I have been to Peter Luger's a few times and it is indeed quite tasty, I just thought Keen's was a notch above.

Second for Keens. Also the pipe collection is really cool
Haven't tried Keens but it's recommended in most of the NYC threads here and on the travel board.
Skip peter Lugers, its overrated and out of the way.
Del friscos, although a chain, is phenomenal. However, I agree with the above poster that i typically try to eat at restaurants unique to a city.

In Manhattan I think it's hard to beat Wolfgangs. He was a maitre'd at Lugers (I think) for many years and now operates several restaurants in Manhattan. The quality of the beef is superb and it's a classic steakhouse experience.

A second for 21 Club. That is another classic NYC joint with beer steins on the ceiling and a secret wine room.
