I love Blazing Saddles and History of the World Part 1 and Big Lebowski and all that, but what I think may be the movie I've laughed the hardest at, and this is going to be sad to admit, was Harold & Kumar: Escape from Guantanamo Bay. Rob Corddry is stupid funny in that movie.

H&K2 gets funnier every time you watch it. Fact.

It's gotten some attention here but deserves more. Honestly, I'd put it #2 behind Holy Grail but I love British humor. Salmon Fishing in Yemen started off very similar to In the Loop but somewhat crashed and burned by the end.

Blazing saddles is a great comedy, wedding crashers, dumb and dumber, and The Big Lebowski are my go to movies for laughs.
