If you watch the clip, you can clearly see that Will Smith is fine with the joke...Jada gave him the look like "2Pac would not have allowed that joke," and caused Will to step up to the plate.

I think the “open relationship” thing is the root of Will’s anger. He can try to pretend like he’s ok with it all he wants, but it’s just human nature to get pissed about your wife sleeping with someone else. He was embarrassed that people knew he didn’t do anything about it and wasn't about to let it look like he just sat by while another man insulted his wife, even if it was just a joke.
