Most likely the colonists that left would never make it in their lifetimes. I would think that there would be several generations born and die out along the journey. Some of whom would never no any other life than being on a spacecraft.

Yes, a wormhole/"warp drive" would be the only way to travel great distances across the galaxy/universe. Just pointing out that it is impossible to travel faster than light.
