To me there are two ways that this question can be viewed: either general accessibility or my personal favorite.

Personal Favorite by Genre

Rock & Roll: Frank Zappa - Camarillo Brillo
Jazz: Bela Fleck & the Flecktones - Poindexter
Hip Hop: Jadakiss - One for the Love
Classical: Beethoven - 9th Symphony (Not a "song" but my pick for the most moving piece of music ever written)
Folk/Country: Leo Kottke - Pamela Brown
Punk: Operation Ivy - Knowledge
Alt Rock: Ween - Light Me Up

As for "Best. Song. Ever" I would probably go for:

Led Zeppelin - Stairway to Heaven

It has been so successful that it is cliche. If you live in the Western World and don't know this song then you just don't listen to music, and you probably live under a rock.

A similar argument can be made for:

Beatles: Hey Jude

And to a lesser extent:

Beatles: A Day in the Life
Queen: Bohemian Rhapsody
