1. Welcome to Jurassic Park
There is not other scene in movie history, IMO, that captures the essence of film making as this one scene does. The cinemetography, the acting, the dialogue, the special effects, the score, the list goes on and on. The way this scene makes me feel every single time I see it is what storytelling is all about. It is truly overwhelming.
2. Introduction of Hans Landa
I can't think of another scene that captured the essence of a character more in one clip than this one. The tension can be cut with a knife it is so palpable.

Someone mentioned the "processing scene" in the Master in another thread. Great scene. The opening to Inglorious Basterds would easily be my number one. Other Tarantino scenes would probably fill up 10 of my top 15-20.
