Well Locke and MIB talked before the statue was destroyed down in the wheel chamber, so he knew Richard was alive in the future, so that may have a lot to do with why he spared him, especially the second time when he didn't kill Jacob.

Seriously doubt it. MIB was on the Beach camp in the Incident and Dr. Linus, plus a few other manifestations throughout the series (Yemi and Boone for example).

Do we know this for sure?
The island jumped right before that conversation. We know it was a point in time prior to the well being built, but I don't remember them showing the whole statue in time frame.
They showed the statute in tact two jumps prior.

It flashed as Locke was going down the well. Locke then hits the ground and breaks his leg. Up on the surface Sawyer and co. see the back of the statue, and then Locke pushes the wheel and they are flashed to 1974.

BTW, if Christian is MIB in early season 1, then he was on the beach and in the ocean.

Just went back and watched it on Netflix. They don't show the statue at all after the flash while Locke is going down the well.
The statue was shown several flashes before.
So we don't know what year it was when MIB/Christian spoke with Locke.

I'm not sure if this is the first time he's manipulated people to get off the island, but I do think that Ab Aeterno was the first time he used a human to try and kill Jacob.

I completely agree, my premise is "this time, we're going to tell you the story of the final time" - kinda going off the Matrix theme - "this has happened many times, but we're going to show you this one, because this time it ends differently than it ever did before"

I'm pumped about seeing John Locke's wedding the in the alt timeline (I assume we will see this at some point). I'm wondering if we will see the same Anthony Cooper as Locke's father or if it is someone else.

Nope, all part of the game.
For the "loophole" to work, Locke has to get the candidates back to the island and he has to be dead.
Obviously it's going to be tough to convince everyone to get back, so the easiest method is to convince the leader, Jack. Locke even references this off island when he tells Abaddon, "I only have to convince one, the rest will follow."
MIB tells Locke "say hello to my son." Locke makes the connection and uses it to convince Jack to come back to the island.
Thus, step two of MIB's loophole is accomplished.
