The Expendables maybe? Not out yet but maybe action-wise might be on that list? Willis, Statham, Stallone, Li...other guys just, whatever..

Mad Mad World..comedians of the mid-century.
Longest Day..my call for #1.
A Bridge Too Far..testosterone stars of the era.
The Departed..modern day stars aplenty.
Godfather 1 and 2..Coppola made relative unknowns.
Apocalypse Now..see above.
The Towering Inferno..1970s polyester=fire hazard!
Gumball Rally..Mad Mad Redux w/cheesy 1970s stars
Saving Private Ryan..3 hr war films = huge casts.
The Player..Altman films, like Scorsese and Kubrick films, always have strong casts.

you need to define this better. just throwing a bunch of big name actors into a movie doesnt mean the movie was great.
i could argue the relative no-names in serenity was a better cast because they interacted so well together.
likewise you have dustin hoffman and warren beatty in Ishtar and its a complete disaster.
something like true romance would probably get my vote..just about every majobr actor who appeared in that movie was used to good effect.

Greatest cast of any movie ?
Are we talking about a movie with many stars or a movie that was cast well?
A movie in the first type:
Grand Hotel 1932
Greta Garbo, John Barrymore, Joan Crawford, Wallace Beery, Lionel Barrymore, etc
Just like It's a Mad Mad Mad Mad World it's an ok movie nothing speical.
Movies of this first type tend to be average on scale, with a way for studio's or todays money makers to be placed in one film to draw a larger audience.
Movies of the second type are more common and in film history stand out...
Casablanca
I can name no one that was missed casted in that movie.
To Kill a Mockingbird
Can you see anyone other than Peck as Atticus Finch or Brock Peters as Tom?
The Searchers
Who would replace John Wayne as Ethan Edwards or Hank Worden as Mose Harper? Look at Ken Curtis' role of Charlie McCorry which could be his best job as an actor ever, or Jeffrey Hunter as Martin. This is a movie it would be very hard to come up with a better replacement for any role.
Here is a movie of the second type most people have not seen:
Dr. Ehrlich's Magic Bullet
Edward G. Robinson plays Dr. Paul Ehrlich, and this well could be Robinson's best acting job ever.

They couldn't afford to make it today. Everyone would want a big slice of the pie.
They did make a smaller toned down semi-version of Mad Mad Mad in Rat Race.
