Laws of Thermodynamics make heat death an inevitable end of the universe. There will be no more energy available for work at that point.

i would think #2 is a bigger question. it ties much more into existential thought & whether there is more to the universe than meets the eye. aliens existing would be cool as shite, but it wouldn't prove or disprove anything about any "greater meaning" or ultimate fate of the universe that we perceive.
fwiw i think people are conceited if they think that humankind perceives all that is going on. i like to look at the universe with the idea that human intellect is fundamentally limited and that there are things we can & will never understand. a good analogy would be that your dog will never understand the extent of human civilization. in this way humans may never grasp the greater organization of the universe.
and so, with that in mind, i choose #2 as the bigger question.
