A big excess of beads with a lot of capture antibodies on them means that your target protein is going to be vacuumed up out of solution really quickly, and as this post explains, that’s a real advantage in these methods. The more time proteins spend rubbing up against and rolling across surfaces, the more degraded and denatured they get, so speed is definitely a virtue here. Once you’ve done this, then (as with an ELISA) you set up a “sandwich” assay format, where your protein of interest ends up captured by one antibody, while its remained exposed surface then gets bound to another detection antibody. This detection antibody has biotin tags already attached to it, and every time you see biotin stuck onto something, you know that it’s going to be used to grab onto some other reagent that has biotin’s lifelong love, streptavidin, attached to it. Indeed, once you’ve hit the sample-incubated beads with the detection antibody, you add in a streptavidin-beta-galactosidase enzyme conjugate, so that in the end, every bead that captured a target protein now has a detection antibody stuck to that in turn, which is then stuck to a streptavidin-enzyme species. A sandwich indeed!

IDGAF if people get it or not... it's their compulsion to loudly and proudly share the patently incorrect reasoning. Unteachable is worse than unvaccinated.

Anyhoo. What's the forecast, Peej?

it's a lab assay that has a cute name (sandwich) to differentiate it from other similar types of assays

here you go:

The spike protein is located on the outside of a coronavirus and is how SARS-CoV-2 (the coronavirus) enters human cells. Its location on the outside of the virus makes it so the immune system can recognize it easily. The spike protein is unique to SARS-CoV-2 – it doesn't look like other proteins your body makes.Jul 2, 2021

No lunch. Still miserable with my assumed to be flu. Sucks. Missing the wine tasting girls night tonight :( And I was supposed to make the desserts, so now I have a metric ton of key limes and other dessert ingredients sitting here.

if you can still taste things and have the energy to do so, I think you should have a weekend pajama dessert binge

I can not. Did you lose them also? I can still taste coffee, a little.

Frozen 'za. Added garlic, oregano, mushrooms, bell peppers, onions, black olives, parmesan, mozz., and Krystal. Coffee, black.

I am seeing some interesting pizzas from south LA being posted occasionally. It looks good, and I may have to give it a shot soon.
