It's too hard to pick just one. Lots of good ones listed already. A few that come to mind, in no order:
There Will Be Blood - So many good scenes; this is probably my favorite: LINK
Drug deal scene from Boogie Nights: LINK
Gas station scene from No Country: LINK
Many scenes to choose from in The Godfather. "It's strictly business" - LINK The Sallazzo kill scene. "I never wanted this for you" - LINK The baptism scene. Etc.
And the best Goodfellas scene, hands down, is the "funny guy" scene: LINK

Such a hard decision, but I love the scene in bad boyz 2 where Will Smith and Martin Lawrence interrogate the tall kid who comes to the door to take Martin Lawrence's kid on a date. 
Also love the scene where Tommy beats down mad dog grimes in warrior.
