The scene in Kingpin where Roy sees McCracken's commercial in which he sponsors fatherless families. All of the moms are MILF's and Roy just sits there in disbelief. That last line of the commercial goes somthing like, "Its a tough world out there, these kids nearly got munsoned but now they're back. Through the Unifed Fund I found out if you give a little, you can get back a whole lot more."

There's a scene in "The Money Pit" where everything in the house goes wrong, and Tom Hanks loses his shite and puts together the most hilarious laugh I've ever seen.
The movie itself is pretty mediocre, but this scene is spectacular and never, ever fails to make me laugh.
