Indy 500 as an individual race. Of course it hasn’t been the same since the split, but as far as one race that means the most to an individual driver’s career, I think winning Indy is it. The event itself is the largest attended sporting event in the world, has tons of tradition, and has experiences for everyone from the blue collar to the wealthy.
Monaco is a terrible race and everyone knows it. F1 has a global audience so they do have crazy amounts of viewership and fans, but Monaco is an event, not a race.
Le Mans is great but sportscar racing just isn’t as big as open wheel.
Daytona means nothing globally but obviously it’s the biggest deal to Americans. NASCAR is on a downhill slide though, and the influence of the sport (and it’s biggest race) seems to be shrinking back to the southeast of the US.
ETA: I am very biased though.

24 Hours of Lemans IMO. No other race has the amount of money spent to win that particular race. The numbers of fans in the "stands" and watching on television is huge.
The GP of Monaco is pretty much a parade, the qualifying does by far the most to set the finishing positions.
The Indy 500 is a shell of its former self.
The Daytona 500 at its height was not a lot more than regional racing and not even close to being in the discussion.

Either Le Mans or Monaco, probably Le Mans over Monaco historically but nowadays its Monaco.
I'd rather go to the Monaco Grand Prix but I'd rather be in the pit for Le Mans.
