Ego's brain was at the center of his planet. He literally explained how he did it when talking to Quill and showed him building layer upon layer on top of it.
Knowhere, as explained by Gamora (assuming she actually knows though it may just be legend) is the SEVERED HEAD of an ancient celestial being...meaning it was severed from a body.
So we'd be talking about a being that was enormous in size...think Galactus.

They've been vague on Knowhere in the MCU. But we also haven't gotten Cosmo running it. Just shout-outs.
Galactus has sizing issues. He's been everything from 30' to fleets of capital ships are dwarfed by his presence.
Usually though he seems to be within spitting distance of the Celestials supposed to be 100'ish statures.

There's an internet theory that Ego originally had a body and was decapitated, leaving the head to become Knowhere. But, being immortal, his brain reformed into "the living planet" with no memories.
