Lambert Field (Purdue's stadium) holds 1,100 people. For comparison, even UCLA's Jackie Robinson Stadium had a crowd of 2,613 when UCLA and LSU met in the winner's bracket of the Los Angeles Regional back in 2010. Here is the criteria for selecting regional sites:
Lambert Field does not have lights, which is the main problem. Not sure if their stadium can generate enough money, either.
They are building a new stadium that is set to open for the beginning of the 2013 season.

The current stadium has seating pretty much just around the batting circle. The new one will have seats past 1st and 3rd base on both sides, a few 2nd level bleachers behind the plate, plus a few boxes. Also, there's the hill down the 3rd base line.
To me, it looks like it'll seat a lot more.

why not play at the minor league park in INdianapolis? 10,000k seats or so vs 40,000
