My top recommendation would be Red Dead Redemption. It's amazing how much fun some of the most mundane tasks are in this game.
If you want to spend 40-60 hours in an experience, check out the Assassin's Creed series. I would recommend getting the detailed story from the internet for the first AC game. It's okay, but extrememly repetitive...you do the same few things over and over again for each mission. It is cool to go around the maps, though. While there's a cliffhanger ending, the main "assassin's" story is self contained.
Then play AC II and follow that with AC: Brotherhood...they are essentially one long story. After Red Dead, these are my two favorite non-sports games of all time. You can easily spend 20 hours in each game...I was closer to 30 on each. Each version improves on the previous one.
Batman AA was another incredible game. I highly recommend this as well. You can beat the hell out of everyone, or you can mess with their minds...which is the most fun way.

If you want a good rental for the campaign only, try Medal of Honor.

Couldn't have said it better.
I would add Uncharted in there to. UC1 was one of my favorite single player games ever. UC2 was good and is a must play, but not as good as 1 IMO.
