Looking for advice on New England

Wife and I are going up for the second week in September. Planning to go to Boston for 2 nights then Martha’s Vineyard for another 2 nights. That’s as far as we’ve gotten. Is 2 nights to many on Martha’s Vineyard?

No two nights should be plenty. I wouldn’t say you need any more than that though.
Personally I’d prefer an extra night in Boston, but that’s just my opinion.

Don't know anything about MV or whether 2 nights might be too many or not enough. If you are going to be in Boston, Portland, ME might be a good option.

Maybe spend one night on Martha's Vineyard and the other night on Nantucket.

That’s a lot of time on ferries.
Unless you are taking Sandpiper Air from island to island.

IMO yes, since the beach weather will be long gone. But two nights is not necessarily a bad call. It takes time and effort to get to/from MV, so two nights makes the ferrying more worth it.
I definitely wouldn't spend more than two nights though. In the absence of beach weather, there's not much to do on MV. It gets pretty dead in the off-season. And it's a pretty small island.
There are a few things I can recommend on MV.
On the Western side of the island, there is a tiny fishing town called Menemsha. There you will find a little seafood shack called Larsen's Fish Market. It is a very unassuming place. You'd have to know it's there. Get the lobster rolls for lunch there one day. It is literally lobster meat on a hot dog bun, but it was the best lobster roll I've ever eaten. I ordered another after I ate the first one.
It's pricey, but if you can swing it, eat dinner at The Terrace at the Charlotte Inn in Edgartown. One of the best meals I've ever had. Also, props to their bartender who made the best Old Fashioned I've ever drank.
For good views, there is the Aquinnah Cliffs overlook on the western end of MV.

Have you thought about going the other direction? Salem, Rockport, Gloucester, Manchester by the Sea?

Saying 'nights' is really an amateur hour term. Because when traveling, 2 nights really means 1 whole day guaranteed and who knows what else. I mean you could leave Boston at 8pm after dinner the first night and leave first thing the 3rd morning. There's going to be a commute to and from there.
I mean 1 night is always crazy. You get there for an evening, spend half the day the following day and then are off. If that's all you need no problem, but I'm saying you can easily spend much more time then one evening and half a day in Martha's Vineyard.
