I'll have to go with Phillip Seymour Hoffman in Along Came Polly. The entire movie is worth watching just for his scenes. He was funny as hell playing those bagpipes, though.

That was a good one....when he decided to take the lead role in Jesus Christ Superstar in the middle of rehearsal, that was hilarious.
