What are the main differences that teams look for when staffing a 3-4 ILB and a 4-3 MLB?

I think it's more dependent on the type of 3-4. Like more of a Zone blitzing 3-4 or just a base. But I'm probably wrong

Since you only have 3 linemen (Nose and two tackles) the Nose needs to be big enough to take up two blockers.
Your rush linebacker is a tweener type linebacker/DE meaning he doesn't have to be 280 lbs like Cam Jordan or Will Smith.

Plus since you are always rushing a LB you can play more games and move him around.

Vilma should be perfectly capable of playing a 3-4 MLB. If not Hawthorne should be capable. If we keep Vilma, those 3 need to be on the field. then you put in your speed guy at the other side.

I think this speaks volumes of what they think of Jordan. This will also get Bunkley on the field way more. He was great on the few snaps he played

In a base 3-4 scheme, a ILB is typically slightly bigger. A 4-3 MLB like Vilma can be protected by two fat DTs. Not so in a 3-4.

You really want guys to be about 6'3" and 240 lbs.

3-4 inside backers have absolutely got to be able to shed blocks and play in a phone booth. It's why Vilma didn't play well at all when the jets put him there so many years ago..he's just not big enough.

In a 3-4, the inside backers generally have responsibilities from Guard to Guard, while in a 4-3 it's from Tackle to Tackle, or sometimes even more than that. Need more speed out of a 4-3 Mike because he's got to be able to roam around the field and go sideline to sideline.
