Does New Vegas pick up where Fallout 3 left off? I haven't played Fallout since the first game so I'm wondering if I'll be lost if I get New Vegas?

New Vegas takes place in Las Vegas and Fallout 3 takes place in Washington DC if that answers your question.
They're both stand alone games with their own cohesive stories, but you're missing out if you don't play Fallout 3 too.

Ok I just didn't know the storyline of the games since I haven't played one in so long.

Thanks for the heads up on the sale. I read on this board that Bethesda games can be buggy, so I held off on New Vegas when it first came out. When I saw there was a major patch/update released recently and the game was only $10, it became a must buy for me.
I picked it up at the Best Buy at the Mall of LA. If anyone else goes, I didn't see it on the shelves, so I had to ask a guy to get one out of the back.
