No one really knows. There is a comic called "The Killing Joke" that goes into how this happened. In the comic he is a shitty stand up comedian whose wife is about to have a baby. Since he isn't making money, he agrees to help some criminals get into an old building. The criminals are going to make him the fall guy, but he doesn't know that. Before he helps them, his wife and baby end up getting electrocuted I think. The bad guys give him a red mask to wear. He then ends up falling into a vat of chemicals as Batman is chasing him. He turns into the Joker. Now I read this comic a while back, so I could be wrong with some things here. Anyway, some people go by that, others don't. I think the point of the Joker is that you aren't supposed to know why he's evil. He just is. He's the perfect nemesis to Batman. BAtman is supposed to pretty much be all good and Joker all bad. Thing is, shades of gray do develop since Batman isn't so different than a lot of the criminals he faces.
