Look at Nasuni. If I recall correctly, you can check out files. Its cloud and on Prem.

How much total data are you storing? How much of it needs to be readily accessible?

We use SharePoint Online for file check in/out. Not sure about check in/out for files that big.

That is quite a can of worms. What do you use now, or what would you use on a regular file server?

Do you really? I ask because VDI may be a solution for you. In a past life we ran CAD from Azure (VDI). I would research VDI for sure.
