All of this stuff.
Also, check out Trastevere, it's a little neighborhood in southern Rome that has an authentic feel. Really awesome riverside neighborhood, it was my (and my wife's) favorite part of Rome.
Remember to pack comfortable shoes.
Trevi Fountain is a must see, too.

1. Vatican Museums/St. Peter's Basilica to Passeto di Borgo to Castel Sant'Angelo
2. Palatine Museum, Roman Forum, and Coliseum (these are often grouped in guided tours)
4. Pantheon/Santa Maria Sopra Minerva/Piazza Navona/Largo Argentina (where Caesar was assassinated)
5. Campidoglio
That's enough for your first trip.
