The best 2 player Genesis game is, hands down, Gunstar Heroes.
Other good 2 player games I haven't seen mentioned:
Zombies Ate My Neighbors (though, it's better on SNES)
Bonanza Brothers (similar to Elevator Action)
Herzog Zwei (If you enjoy strategy games)
A couple of more obscure single player games:
Beyond Oasis (Sega's answer to Zelda)
Flashback: The Quest for Identity
Shinobi 3
Earthworm Jim
Aladdin (Yes, based on the Disney movie)
