The mother's mother was an old crone of some sort that people used to come to for potions and elixirs....particularly love potions.
The books note that Tywin had the mother give Jeyne "moon tea" to prevent a pregnancy. It is not a huge leep to assume the mother gave each of them a love potion as Jeyne was nursing Robb back to health after his injuries in the west.
So yes, it is my belief that Tywin sowed the seeds for the Red Wedding months earlier when he had Robb and Jeyne given love potions by her mother. I believe he just seized opportunity when the Freys and Boltons agreed to set up Robb at the Frey - Tully wedding.

AKA Maggy the Frog who gave Cersei her Valonqar prophecy. And yeah, pretty sure that's how it went down.
