Round Up and similar herbicides work on live foliage. As soon as you see newly sprouted leaves (groups of 3), spray them. The actual time intervals will vary with heat and rain.
You might try digging out some of the roots. They run horizontally just a couple of inches under the surface. Two problems to that approach are- (1) the roots are also toxic, (2) they look like most other vine roots.
