Here's a really deserved nomination that I haven't seen mentioned.
Rulon Ellis Gardner won the gold medal at the 2000 Olympic Games by defeating Russia's three-time reigning Olympic gold medalist Aleksandr Karelin.
Rulon wasn't just an underdog. Karelin was possibly the most dominant athlete ever prior to losing. Karelin had been unbeaten for 13 years in international competition prior to the upset.
