How do they know who is a bot and who isn’t? How do they know what to look for? A certain algorithm or something?

That's what I want to know.

I've seen some (assumed) bots on YouTube comments. They create a user name similar to some popular channel owner, use the channels picture as theirs, and then offer a 'gift' to another commenter, with a link. Of course the 'gift' is some sort of scam that will require $ to obtain (shipping, etc).

Those are easy - you go to the scammers profile, and there is no info there, just these repeated same scam posts. Not sure how you could tell with a poll - maybe they can easily and automatically get a profile of the poll voter's content. If it fits a certain profile (reposting the same spammy stuff over and over, etc), it's labelled a 'bot'?
