That’s a tough one. How does one judge a “star”? Heights of fame? Longevity? A combination?
Eddie Murphy was probably the biggest star in terms of reaching the heights of stardom. Dude had box office blockbusters, standup stardom and even a song go platinum (album went gold).
Bill Murray likely has had the longest career, but even with ghostbusters, I don’t think he was ever as popular overall as Murphy at one time. He’s basically a folk hero at this point.
Sandler is kind of that combo. Obviously not as much longevity as Murray yet, but he’s had more star power than Murray, but not as much as Murphy did for that time period in the 80s.
With all due respect to Dan Ackroyd and Mike Myers, I’d have to say it’s either Murray, Murphy or Sandler.

Robert Downey Jr didn't get his start on SNL. He was on the cast when they intentionally brought in known names to help boost the ratings. This is a general response to the thread and not necessarily the poster whose post I responded to.

Yes, Billy Crystal and Martin Short also fall into this category. Both of them had been on TV shows or variety shows and had bit parts in movies before being on SNL.
