Jesse suspected huel of jacking the ricin and confronted Walt at gunpoint about it (think it was end times episode in season 4).
But Walt was to able to bs his way out of it.
Now that it happened again. Too much coincidence. Though I think Jesse was looking for his weed and it was gone.

It doesn't make sense for Jesse to look at his cigarettes and figure out the whole Brock thing. Either Walt or Huel switched his pack for one laced with ricin and he realized that the # of smokes was off.

Jesse didn't have cigarettes on him, he had weed. He realized that Huel switched the two and then he connected the dots on how the ricin cigarette disappeared.
