Try to set up a guided tour through the Vasari Corridor. It will take you from the Uffizi all the way to the Boboli Gardens and Palazzo Pitti.
The Accademia to see Michelangelo's David was mentioned earlier in the thread. If you can get there early to beat the crowds or book a line-skipping tour, then I'm all for it. Otherwise the lines are ridiculous and it is the only real must-see in the building.
Go to Sienna as a day trip.
I see that Basilica di Santa Croce was also mentioned. Can't skip seeing where a Ninja Turtle was buried. Speaking of Ninja Turtles, Donatello is also buried in Florence in the Basilica di San Lorenzo.
