My day job is actually Anthropological Animalogist, so I've been studying terms like these for years.
How this one starts is actually a neat story. So, back before farmers ear tagged their livestock, it was hard to keep count of their bovines. Once the "storing room" was filled after the day's slaughtering (a total of 246, generally), they could count how many were left over in the barns and fields.
Thus, that how it came to be known as "A Murder Of Cows"
