No, as the last info I have you are not allowed to set foot on the islands. There really is no one there to enforce it but that is the law do to bird nesting. LINK

The Chandeleurs are my favorite place to fish when I have the time for the run from Biloxi, and yes the trout fishing there can be great.

There are several outfits that run charters down from the MS coast both day and overnight LINK LINK among others.
as well as a barge that is generally there with overnight accommodations LINK

As far as time of year, that would depend on your target species, I have caught cobia out front of the islands all summer, flounder are pretty good spring and fall, redfish pretty much year round and trout in the spring and summer. I can't speak to the winter as I avoid being wet and cold in a boat offshore as much as possible and spend my time in deer stands and duck blinds.
