I love discussions like this, because they are so subjective, and not sure if their is a definitively correct answer. Glad Les Paul was mentioned, because he really brought the guitar to the forefront of popular music. Before him (with some exception - especially in C&W music), the guitar was just a part of the assemblage of instruments.
It's hard to make an argument against Jimi as the best ever, but most influential...I don't think so. After you get past Les Paul, these two bands had multiple people who influenced rock n'roll gyitar playing: The Yardbirds and John Mayall's Blues Breakers.
