Everything I've read so far as hunting says live capture by hand, noose, trap.
As far as transporting them, I'm in error since it is illegal to transport live pythons by vehicle---again, according to what I've read.
Once captured, they can be dispatched by the hunter by destroying what little brain they have either by knife, some other sharp implement or by gun.
