Bioshock - the big reveal about your benefactor's real identity and his mean's of directing your efforts. Still enjoy a playthrough every now and then, but you only get to experience that reveal once.
Armored Core 1 - was fun to try and fail until you learned all the quirks and best equipment and mission strategies.
The second one goes for a lot of the great games played. First playthroughs have a bit of magic, if you will, because you don't know what's going on. FF7, FFIX, RE1, RE2, LoK:SR, BoF3, Medevil, Onimusha: Warlords, Borderlands 2, Diablo, Diablo II, Darkest Dungeon, Nobunaga's Ambition:Iron Triangle were the most memorable for me. The figuring it out is the best part.
