The Panama Canal seems like the obvious thing to do, but I have read the city has quite a lot to offer besides that.
Spent a week down there a few years ago on business. Went a couple days early so I could take in the sights. We hired a local guide to take us around. I highly recommend you do the same. We spent a day with him seeing the city, canal, and even out in to surrounding countryside. Another benefit of having a local guide is he knows what areas you don't need to go to.
