The opening seen looks like it took place on Earth. If you stick to the movie's angle, that the Engineers created humans and the quote about you must first destroy in order to create ( or something like that), you can assume that mankind was created when the Engineer took a sip of the black goo and fell into the water.
What doesn't add up is how there was a 100% DNA match if his DNA was ultimately broken down, then rebuilds in the water?
