I was listening to NPR today, and they asked a similar question:
If JFK had been similarly baselessly accused of being a Russian agent,
would the Cuban Missile Crisis have ended differently?

The theory was that these baseless accusations have de-legitimized the presidency, and that the only way for Trump to disprove them was to launch the strike on Assad in defiance of the allegations that he's a Russian agent. This has completely destroyed any chances for detente with the Russian government.

If Kennedy had been saddled with similar baseless accusations weighing down his presidency, clouding its legitimacy, he would have had zero choice but to launch a strike.

Unlikely. The Russians responded to a show of strength, appropriately, by backing down 90 miles from our shores.

Weakness emboldens aggressors.

On the other hand, what would have happened if a dilettante like Clinton or, worse, Obama had been in charge? "Better Red than Dead" would have likely led us to capitulation.

Yes there would have been war, and Trump would have consumed those Russians with lightning bolts from his eyes and firebombs from his arse!
