To keep on the nightstand the best value out there is a 6" GP100 .357 mag. With full power .357's it's about as much horsepower as you can get in a handgun that's still portable and reasonably easy to shoot.
Smith and Wesson has better self defense offerings (and they are more refined guns) but they cost a bit more and have hammer block safeties that can potentially fail, but failures are extremely rare. A 6" smith 686+ gives you 7 shots in a very nice revolver. The R8 is the ultimate in self defense revolvers with 8 shots, moon clips, and a fantastic trigger. The downside is again price but it's arguably one of the best self defense handguns a person can own if concealability isn't a concern.

I had been wanting a revolver too. Last year I came across two S&W 686s, one a 686-1 6" and a 686-6 + 2.5". I couldn't decide so I got them both. Sweet guns, both now my favorite. The big one shoots so incredibly soft.
If I were you I'd look for a 686+ 4". Perfect nightstand size.
