Definitely tough. It may be the people I know or whatever, but I feel like Sam Cooke is vastly underrated. He is so smooth - could break your heart singing a Waffle House menu.
ETA: Of these three specific choices:
Sam Cooke
Otis Redding
James Brown

I've always found Redding somewhat overrated
Brown was the greater showman, but for singing it's clearly Cooke
Please forgive the plug:

I can see it, but I think he is too often just "Dock of the Bay" Redding, and not enough "Try a Little Tenderness" or "I've Been Loving You Too Long" Redding.

There are no wrong answers in this thread.
Pain in My Heart - Otis Redding
Please, Please, Please - James Brown
(I believe Ric Flair learned some of his routine from this vid.)

"Wicked" Wilson Pickett
James Brown
Sam Cooke
Otis Redding
I like my soul music gritty and funky. I get goosebumps everytime I hear Wilson Pickett's version of "Hey Jude."

Sam Cooke & James Brown is tough to chose.

The Cooke and Redding comparison is a fair one, but, in my opinion, the Godfather of Soul label bestowed upon Brown was unfitting. To me, he performed more of what eventually became funk rather than legitimate soul.
Cooke predated Redding by a few years and therefore might be the more appropriate King/Godfather of Soul. Still, Redding's death at such a young age makes you wonder about what might have been.
Cooke certainly had the cleaner vocals, but Redding had so much more raw emotion behind his voice.

Cooke .. no doubt IMHO
Redding was more narrow in his scope
Brown was a showman

It's easily #1. Tramp is his best song.
