Comedians in Serious Roles

What is the best serious movie role that was cast with an actor or actress that typically does comedy? To make this a bit more difficult...I will not include Robin Williams in Good Will Hunting (low hanging fruit). Outside of that role...who do you think took a risk in casting and hit it out of the park?
Mine is a bit off the wall, but I thought Bill Murray did a great job in The Razor's Edge.

any Tom Hanks role in the past 20 years?
People forget he was of Bachelor Party, Bosom Buddies, etc.
I know he did some stand-up, but that probably was never his "career" and he may never have been defined as a comedian so not sure if it counts.
