I generally avoid St. Pete as its out of my way but if you're willing to do Tampa I'd suggest the Columbia in Ybor, Bern's Steakhouse, the Tampa Taco Bus, Yummy House, and the Salt Rock Grill as all good choices.
As far as musts go, the Columbia Ybor in my opinion is the defining Tampa restaurant from both a culinary and cultural standpoint.

If you enjoy middle eastern cuisine there is a Persian place in Clearwater called mirage. If your not familiar with Persian cuisine it is although very similar to other middle eastern far more complex.

Chicken tadik, a Persian dish that is awesome. Rice cooked with saffron, raisins, onion, seasoned boiled deboned chicken meat prepared a la a layered casserole(lasagna) that is poured into a bed of a yogurt/egg/butter bottom. The liquid goes in first, then a layer of rice followed by the other ingredients, and then a bit of the liquid is poured along the edges of the casserole dish. When it is baked the liqiud cooks as if a crust. It is then served in the manner of an upside down cake, and properly done eaten with your fingers, and other fruits and veggies.
