Thinking about heading up there in Oct for a long weekend. Anyone have suggestions on what to do and where to stay?

There are amazing breweries in Asheville. The brand new Sierra Nevada is insanely nice, as is the new New Belgium. There are obviously tons of smaller ones as well worth checking out.
Here are some shots of Sierra Nevada:

Search Asheville on this board. There was a thread a few days ago and a couple more over the past few months.
The Sierra Nevada Brewery is amazing. I felt like Charlie in the Chocolate Factory. I heard a tour guide say you have to book the tours a month or so in advance because they're so busy. You can do a self guided tour through a overhead corridor that stretches through the brewery. The restaurant at the brewery is good too.

Another vote for Sierra Nevada..fantastic venue and good beer and food. New Belgium just opened up their tasting room, haven't been yet.
I'm partial to Wicked Weed because they make two of my favorite beers (Freak of Nature DIPA and Pernicious IPA) and I like their bar food.
I usually eat at Salsa's for lunch..it isn't Mexican or Tex-Mex, but a take on stuff like burritos, etc. with fresh ingredients. I think the chef is Cuban.
Plenty of good dining options for dinner..hard to suggest without knowing your taste.

Head up there there in mid-late October when the leafs change. Get a room
At the downtown inn, Drive the Blue ridge parkway, get out your car take pics and hike. Lots of breweries and great food downtown. I was there this past weekend and had a filet of Carolina bison from the Market Place, delicious and exotic to say the least.

I went back in January, and I'm heading back this weekend. Sierra Nevada is definitely worth seeing. It's beautiful.
I'd throw in Wicked Weed, Funkatorium, and Burial as must visits for breweries. Hi-Wire and Wedge are very good as well.
As far as food goes, I really enjoyed Biscuit Head and White Duck Taco Shop.

Pisgah brewery has outdoor concerts every so often
Curate for dinner restaurant- get reservations
Farm burger for lunch
Food tour or beer bus tour
Off the wagon piano bar is fun if it's busy
Land Rover driving school if you got money to blow
Hike and see the true "gods country"
Agree with many of the above

I'd recommend spending some time in West Asheville. A little more laid back than downtown. Hit Altamont for beer and Universal Joint for a burger.
