Where are you from and what are your interests?
Disneyland or universal would be fine if you have never been to a theme park.
A tour is just going to drive through Hollywood and Beverly Hills... Hollywood is a disgusting tourist trap but Beverly Hills is obviously a nice area, especially if you're curious to see what a $50m house looks like.
Keep in mind LA weather is awesome even in October so you could consider visiting any of our million beach communities, i.e. Malibu, Santa Monica, etc.
If you are curious to see Hollywood I'd recommend Venice beach instead. Its adjacent to Santa Monica, and features the same freak shows your going to find on Hollywood Blvd, but at least you will be at the beach. Venice is pretty lively on weekends, not sure about weekdays though.
If you're wife is into shopping there's rodeo drive.
I live in LA and I love it because there's so much variety and so much to do, but I really don't see the appeal for visitors to come here.... There isn't really one must-see attraction, and places like Hollywood, universal and Disneyland are giant tourist traps. I guess if you have never been to a big city it's interesting; but if that was the case New York is a million times better to visit.

You could just rent a car and drive around on the interstates. Get stuck in traffic for hours, would be about the same as waiting in line for hours at disney.

LA is so spread out and traffic getting around even on the surface streets can be difficult.
A theme park in LA is not at the top of my list.
There is a lot more to do.
Hollywood Blvd is a tourist trap.
You can get lost driving up trying to find the Hollywood sign but you will end up going through a neighborhood that can help you find the best place to view it.
We walked down Rodeo and other areas.

Head to Santa Monica and Venice. Both really cool places to explore/eat/drink.

Here's the best touristy things I've done in the LA area:
La Brea Tar Pits Museum
Reagan Presidential Library
Nixon Presidential Library
San Clemente (in general)
Santa Barbara (in general)
Santa Monica people watching
Dodgers Game
Getty Museum just the setting and having a nice meal there, I'm not much on art.
I'd still like to go to a game at the Coliseum.

I've got basically the same dilemma on an overnight layover at LAX. We got a room at a nearby hotel and plan on taking an Uber to Manhattan or Santa Monica Beach for the afternoon/evening.

I lived in LA back in 1992 and I've been there just recently. My favorite activity was driving to Point Vicente Lighthouse at Rancho Palos Verdes. It's a stunningly beautiful place but for some reason underrated. I don't see much people talking about it at Tripadvisor or other tourist websites.
Worst was Hollywood Walk of fame area. But it's a tourist checklist thing to do. You'll go there once just to see but probably won't come back when you're back in LA.

RPV is residential so thats why its not discussed much.
Best bet is venice, santa monica and rodeo. For your own sake, skip hollywood.
