It won't happen for a number of reasons. Number one being Uncle Jerrah markets the Cowboys heavily to the SA market.

If the Chargers move, it will be to L.A., not to San Antonio. Personally, I think they should stay in San Diego and the Jacksonville Jaguars should move to L.A., but that's me.

Rick Perry should research San Antonio's sports market before he runs his mouth, but then again we're talking about Rick Perry.
The Alamodome would have to be leveled and rebuilt. It was built in the 1990s just prior to the modern stadium boom and lacks in modern amenities necessary for an NFL franchise.
San Antonio may also be a massive and continuously growing city (7th in population), but is a horrible TV Market given its size. The city keeps expanding, but there aren't many large suburbs surrounding the area.
As someone above stated, the Cowboys have a huge market share in the San Antonio. The city ranks second highest in TV viewership of Cowboys games. That's a massive loss if Jerry loses the geographical TV rights to a new franchise.

And didn't Al Davis establish an NFL owner can move a team anywhere he wants?

The Austin-San Marcos-San Antonio metroplex can and would support an NFL team. I think MLB is more likely though.

With NFL's regional coverage, a San Antonio team would more than likely cut out several Texans and Cowboys Sunday afternoon broadcasts on Fox or CBS in San Antonio. San Antonio gets Cowboys and Texans broadcasts during the 11 AM and 3 PM CT time slots. The chart shows that the Cowboys have far more to lose in TV ratings/exposure if San Antonio were to get a franchise (assuming that team would get a normal geographic broadcast region).
Even as I-35 cities like New Braunfels, San Marcos, Kyle-Buda expand, it would take quite awhile before Austin and San Antonio merge into one TV market.

Like 10 years max? I drive it quite a bit and San Antonio and Austin are rather quickly getting to be quite contiguous.
It will be pretty interesting to see which major sports league comes to Austin/San Antonio next - two decent sized markets destined to become a top ten metroplex...
