Technically, the heirs to the Baratheon throne are:
Stannis
Aegon Targeryen
After that, the line of succession is dead. And I don’t think anyone is going to rally to a Lannister cousin.
Therefore, since we are pretty certain that Tommen dies, the next question would be:
Who is in the best position to take the Iron Throne and the power to hold it?
1. Cersei Lannister - She will not simply step aside and could now assemble all the might of House Lannister. I believe that this will actually happen in the next book and will be Cersei's downfall.
2. Aegon Targeryen – With his growing armies and soon-to-be alliance with Dorne, Aegon is in position to stroll right into Kings Landing as the rightful heir; especially if Dorne brokers a marriage.
3. Mace Tyrell – He has the might and, just as importantly, he has all the key players in position around Kings Landing. He might also get House Lannister’s support if he can get Cersei out of the way.
