I looked in attic to see if ductwork not insulated, can't find anything. Tight. Plenum 12ish ft. upstream is not insulated. That the cause, or something else? If plenum, should I be worried about mold?

Make sure there are no voids in the attic insulation around the duct where the register is. We had that happen. AC man told us that was the cause. Insulated the bare spots and the problem disappeared.

Don’t worry about the mold. Some mildew may form which can easily be cleaned off.

Check the a/c filter. It could need changing. I had a similar problem after installing a new filter that was too restrictive.

They fail over time. Had one that had to be replaced recently. Activated my inner “ I can do Sheetrock repair” guy again. Cost me about $400.

The plenum has insulation attached on the inside of the “box” - that’s likely not the issue - though I did watch one HVAC tech Y/T video some time back where the part of insulation inside the plenum had separated from the box and it did cause some sweating (leakage) at supply registers.

Check carefully around the boot of the supply register (ceiling side and room side) for any leaks or lack of insulation to make sure it’s properly sealed.

Is the ductwork metal Joval round pipe with the wrap around insulation that is stapled closed?

If so, it may be pulled too tight to offer good insulation and the metal pipe is sweating. Feel along the underside of the duct run to see if you find any moisture in that area on the foil back of the insulation.

That's all I've got.

This!! I had same problem. Moved insulation and found water on top of register box. New zip tie and spray foam insulation the connection up the duct about 4”. Have now been doing everyone because I feel the AC leaking around the connection.
