Have two work trips in back to back weeks in these random cities.
Any idea on what to do in either?

Greenville, SC you will want to spend your time downtown. Good local craft beer and chef driven food & nice bars.
Tons of hiking trails within 30 minutes of town. Lots of little rivers and lakes with easy access.

Well you'll only be about 45 minutes away from your namesake.

You'll have more fun in Des Moines than you might think.
What days are you there? How long? Weekend bar scene downtown on Court Ave is pretty good. Hessen Haus is my favorite bar. Get a boot. GREAT restaurant scene, arguably one of the most up and coming in the country. Sculpture garden is pretty cool, go to the Farmer's Market Saturday morning - again one of the best in the country. Exile Brewing Company and Confulence Brewing Company are both excellent - try the Des Moines IPA at confluence. Jasper Winery is underrated. If you like to golf checkout A H Blank. See if there are any concerts at Wells Fargo Arena, usually gets some big names. If you've got a car and like scenery drive south to Winterset. John Wayne Birthplace/Museum and the scenic highways are pretty especially this time of year.

Damn Clemsoners sure admire LSU.
I could spend a couple of days in Des Moines, according to friends who've been there.

In Nashville now but lived there for a lot of years. Other than the winter in Iowa there's not a lot of downsides. I truly loved Des Moines

Des Moines is a cool city. There's been a quick-rise in startups.
As someone else said, the bar-scene on Court Ave is solid.
Other than that, I would go to the Farmers Market (if you're there on a Saturday) and check-out the sculpture garden.

I used to go to Des Moines pretty frequently about 15 years ago. Not sure how it is now, but West Des Moines was significantly better than East Des Moines. I haven't been there since...but I still know people who do. I've heard 801 Chophouse is a really good place to eat (a bit pricey). You may also want to check out Skip's.
As for Greenville...check out The Lazy Goat.
